Enhanced potency of aggregation inhibitors mediated by liquid condensates

Liquid condensates are membraneless organelles that form via phase separation in living cells. These provide unique heterogeneous environments have much potential regulating a range of biochemical processes from gene expression to filamentous protein aggregation---a process linked Alzheimer's and Parkinson's diseases. Here we theoretically study the physical interplay between aggregation, its inhibition, liquid-liquid separation. Our key finding is action aggregation inhibitors can be strongly enhanced by liquid condensates. The mechanism this enhancement relies on partitioning colocalization with their targets inside condensate. theory uncovers how physicochemical properties used modulate inhibitor potency, experimentally testable conditions under which drug potency maximal. findings suggest design principles for respect phase-separation properties.
